A homogeneous split-luciferase assay for rapid and sensitive detection of anti-SARS CoV-2 antibodies

Abstract Better diagnostic tools are needed to combat the ongoing COVID-19 pandemic. Here, meet this urgent demand, we report a homogeneous immunoassay detect IgG antibodies against SARS-CoV-2. This serological assay, called SATiN, is based on tri-part Nanoluciferase (tNLuc) approach, in which spike protein of SARS-CoV-2 and G, fused respectively two different tNLuc tags, used as antibody probes. Target engagement probes allows reconstitution functional luciferase presence third component. The assay performed directly liquid phase patient sera enables rapid, quantitative low-cost detection. We show that SATiN has similar sensitivity ELISA, its readouts consistent with various neutralizing assays. proof-of-principle study suggests potential applications diagnostics, well disease vaccination management.
